    Crossroads at sea: Escalating conflict in a marine protected area in Malta

    This article illustrates how the creation of a Marine Protected Area (MPA) in Malta is failing to adequately include stakeholders in the configuration of conservation targets and measures, leaving local fishers increasingly disempowered. Through a series of interviews and long-term participatory observation, it has been found that the leaders who represent local fishers are failing to communicate the MPA process to their community. Instead, they are using their position in the MPA negotiations to subjugate and silence the fishing community in general and trammel netters in particular. Moreover, in their support for the MPA, these community leaders reproduce the state's conservation discourse to pressure authorities to ban trammel net fishing, with whom they tend to be in competition. It is concluded that the state's narrow focus on ecology, the tight deadlines set out in the EU Habitats Directive, and the misrepresentation of the fishers, has characterised the process of creating this MPA. If artisanal livelihoods are not protected by conservation policies, fishers may regard conservation as a threat to their way of life, and resist policy measures. This compromises conversation efforts and can make the enforcement of the MPAs more expensive. This paper recommends a revision of the community consultation policies of the MPA to allow broader and more representative participation from the local community by encouraging engagement throughout the process as part of a consensual approach to effective marine conservation

    A differential role for the posterior cerebellum in the adaptive control of convergence eye movements

    Introduction The vergence oculomotor system possesses two robust adaptive mechanisms; a fast “dynamic” and a slow “tonic” system that are both vital for single, clear and comfortable binocular vision. The neural substrates underlying these vergence adaptive mechanisms in humans is unclear. Methods We investigated the role of the posterior cerebellum in convergence adaptation using inhibitory continuous theta-burst repetitive transcranial magnetic stimulation (cTBS) within a double-blind, sham controlled design while eye movements were recorded at 250hz via infrared oculography. Results In a preliminary experiment we validated our stimulation protocols by reproducing results from previous work on saccadic adaptation during the classic double-step adaptive shortening paradigm. Following this, across a series of three separate experiments we observed a clear dissociation in the effect of cTBS on convergence adaptation. Dynamic adaptation was substantially reduced while tonic adaptation was unaffected. Baseline dynamic fusional vergence response were also unaffected by stimulation. Conclusions These results indicate a differential role for the posterior cerebellum in the adaptive control of convergence eye movements and provide initial evidence that repetitive transcranial magnetic stimulation is a viable tool to investigate the neurophysiology of vergence control. The results are discussed in the context of the current models of implicit motor adaptation of vergence and their application to clinical populations and technology design in virtual and augmented head mounted display architectures. Significance statement The cerebellum plays a critical role in the adaptive control of motor systems. Vergence eye movements shift our gaze in depth allowing us to see in 3D and exhibit two distinct adaptive mechanisms that are engaged under a range of conditions including reading, wearing head-mounted displays and using a new spectacle prescription. It is unclear what role the cerebellum plays in these adaptive mechanisms. To answer this, we temporarily disrupted the function of the posterior cerebellum using non-invasive brain stimulation and report impairment of only one adaptive mechanism, providing evidence for neural compartmentalization.     As the knowledge base and importance of mitochondrial physiology to evolution, health and disease expands, the necessity for harmonizing the terminology concerning mitochondrial respiratory states and rates has become increasingly apparent. The chemiosmotic theory establishes the mechanism of energy transformation and coupling in oxidative phosphorylation. The unifying concept of the protonmotive force provides the framework for developing a consistent theoretical foundation of mitochondrial physiology and bioenergetics. We follow the latest SI guidelines and those of the International Union of Pure and Applied Chemistry (IUPAC) on terminology in physical chemistry, extended by considerations of open systems and thermodynamics of irreversible processes. The concept-driven constructive terminology incorporates the meaning of each quantity and aligns concepts and symbols with the nomenclature of classical bioenergetics. We endeavour to provide a balanced view of mitochondrial respiratory control and a critical discussion on reporting data of mitochondrial respiration in terms of metabolic flows and fluxes. Uniform standards for evaluation of respiratory states and rates will ultimately contribute to reproducibility between laboratories and thus support the development of data repositories of mitochondrial respiratory function in species, tissues, and cells.     Guidelines for the use and interpretation of assays for monitoring autophagy (3rd edition)

    In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. For example, a key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process versus those that measure fl ux through the autophagy pathway (i.e., the complete process including the amount and rate of cargo sequestered and degraded). In particular, a block in macroautophagy that results in autophagosome accumulation must be differentiated from stimuli that increase autophagic activity, defi ned as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (inmost higher eukaryotes and some protists such as Dictyostelium ) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the fi eld understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. It is worth emphasizing here that lysosomal digestion is a stage of autophagy and evaluating its competence is a crucial part of the evaluation of autophagic flux, or complete autophagy. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. Along these lines, because of the potential for pleiotropic effects due to blocking autophagy through genetic manipulation it is imperative to delete or knock down more than one autophagy-related gene. In addition, some individual Atg proteins, or groups of proteins, are involved in other cellular pathways so not all Atg proteins can be used as a specific marker for an autophagic process. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field

    Por que os meninos são mais indicados do que as meninas como alunos com necessidades educacionais especiais por seus professores (proporção de 2/3 contra 1/3)? Este estudo teve por objetivo comparar a descrição de professores de meninos e meninas identificados por eles como alunos com necessidades educacionais especiais. Os dados foram coletados a partir da coleção de 351 cadastros de alunos identificados como especiais. Esses cadastros fazem parte de um banco de dados iniciado em 2001 por um grupo de pesquisa e contém descrições espontâneas dos professores sobre as características das crianças que, segundo eles, justificavam a notificação. Tais descrições foram divididas em unidades de conteúdo e categorizadas. A freqüência em cada uma das categorias foi computada para os grupos, que foram divididos em função do gênero. Dos 351 cadastros, 66,1% referiam-se a crianças do sexo masculino, enquanto 33,9% eram de crianças do sexo feminino. A comparação da incidência das categorias nos dois grupos apontou como única diferença estatisticamente significativa: a indicação de meninos com base na categoria Problemas de Comportamento. Teorias explicativas existentes na literatura sobre viés de gênero na elegibilidade de meninos e meninas pelos seus respectivos professores são discutidas, bem como são apontadas as implicações científicas e a necessidade de mecanismos de monitoração deste tipo de viés que vem sendo sustentado pela área de Educação Especial e que contribuem para a construção de desigualdades sociais.<br>Why are more boys than girls regarded by their teachers as being students with special needs (2 out of 3 are boys)? This study aimed to compare teacher descriptions of boys and girls identified as students with special needs. Data were collected from the collection of 351 records of students identified by their teachers as special students. The records are part of a database initiated in 2001 by a research group. They contain spontaneous descriptions provided by teachers of student characteristics that could justify the notification. The descriptions were divided into content units and were categorized. The frequency for each category was computed for the groups, which were divided according to gender. Of the 351 records, 66.1% referred to male students, while 33.9% were girls. The comparison of the incidence of categories between groups showed that the only significant difference was the indication of boys based on the category of behavior problems. Based on the literature, the paper discusses theories explaining gender bias on the eligibility of boys and girls by their teachers, as well as the scientific implications and the need for mechanisms to monitor this type of bias.     Safety and tolerability of subcutaneous trastuzumab for the adjuvant treatment of human epidermal growth factor receptor 2-positive early breast cancer: SafeHer phase III study's primary analysis of 2573 patients

    Aim To assess the safety and tolerability of adjuvant subcutaneous trastuzumab (Herceptin® SC, H SC), delivered from an H SC Vial via hand-held syringe (Cohort A) or single-use injection device (Cohort B), with or without chemotherapy, for human epidermal growth factor receptor 2 (HER2)-positive stage I to IIIC early breast cancer (EBC) in the phase III SafeHer study (NCT01566721). Methods Patients received 600 mg fixed-dose H SC every 3 weeks for 18 cycles. The chemotherapy partner was at the investigators' discretion (H SC monotherapy was limited to ≤10% of the population). Data from the first H SC dose until 28 days (plus a 5-day window) after the last dose are presented. Results are descriptive. Results In the overall population, 2282/2573 patients (88.7%) experienced adverse events (AEs). Of the above, 128 (5.0%) patients experienced AEs leading to study drug discontinuation; 596 (23.2%) experienced grade ≥ 3 AEs and 326 (12.7%) experienced serious AEs. Grade ≥ 3 cardiac disorders were reported in 24 patients (0.9%), including congestive heart failure in eight (0.3%). As expected, the AE rates varied according to the timing of chemotherapy in both cohorts, with higher rates in concurrent versus sequential chemotherapy subgroups. In the concurrent chemotherapy subgroup, AEs were more common during the actual period of concurrent chemotherapy compared with the period when patients did not receive concurrent chemotherapy. Conclusion SafeHer confirms the safety and tolerability of the H SC 600 mg fixed dose for 1 year (every 3 weeks for 18 cycles) as adjuvant therapy with concurrent or sequential chemotherapy for HER2-positive EBC. These primary analysis results are consistent with the known safety profile for intravenous H and H SC

    Open data from the first and second observing runs of Advanced LIGO and Advanced Virgo

    Advanced LIGO and Advanced Virgo are monitoring the sky and collecting gravitational-wave strain data with sufficient sensitivity to detect signals routinely. In this paper we describe the data recorded by these instruments during their first and second observing runs. The main data products are gravitational-wave strain time series sampled at 16384 Hz. The datasets that include this strain measurement can be freely accessed through the Gravitational Wave Open Science Center at http://gw-openscience.org, together with data-quality information essential for the analysis of LIGO and Virgo data, documentation, tutorials, and supporting software
